Abstract:
Understanding how the brain functions requires investigating electrical and chemical activities across time- and length-scales. Electrical computation and communication occur in milliseconds, while biochemical signals regulate neuronal function over minutes to days. Furthermore, how these signals propagate spatially within individual neurons, between neurons, or throughout the brain relates directly to how the nervous system processes information and adapts over time. To track electrical signals across time and space, we developed the ASAP family of fluorescent voltage indicators with high brightness, fast kinetics, wide dynamic range, and both one- and two-photon compatibility. Our latest ASAP5 and ASAP6 enable single-millivolt and single-millisecond recording, matching patch-clamp physiology for many purposes, at any point in a neuron or circuit. At the other end of the spatiotemporal range, we developed generalizable bioluminescent reporting methods to provide non-invasive readouts of biochemical activities over seconds to hours. Finally we will discuss new ideas for spatially regulating protein function in vivo for long-term control of network function.
